Abstract

Canadian Pharmaceutical Scientists have a rich history of ground-breaking research in pharmacokinetics and drug metabolism undertaken throughout its Pharmacy and Medical Schools and within the Pharmaceutical and biotechnology industry. The principle of drug Absorption, Distribution, Metabolism and Excretion (ADME) is the foundational basis of rationale drug-design, and pharmacotherapy. The study of ADME and its descriptive quantitative analysis is the basis of pharmacokinetics. Pharmacokinetics is fundamental in the development of a new chemical entity into a marketable product and is essential in understanding the bioavailability, bioequivalence and biosimilarities of drugs. Pharmacokinetics and drug development studies facilitate an understanding of organ-based functionality. Population pharmacokinetic variability and the modeling of drug concentrations has significant utility in translating individual response in a target patient population.This special issue serves to highlight and capture the contemporary progress and current landscape of pharmacokinetics and drug metabolism within the prevailing Canadian context. We invite articles on all aspects of Pharmacokinetics and Drug Metabolism studies highlighting the world-class research currently undertaken in Canada for this special issue.

Abstract

Diet and nutrition are key tools in promoting health and reducing the comorbidities of chronic diseases. There are thousands of biomolecules in fruits, vegetables, wild and medicinal plants, other land and marine organisms, which can exert functional and health-promoting effects through bioactivity beyond nutrition. From the enormous amount of knowledge generated from different natural bioactive ingredients present in foods, we are aiming to bring together experts working in different fields of food, nutrition, and health, in order to work on this Special Issue, with a comprehensive collection of papers to gain insight into the most promising bioactive compounds in different foods, to improve the preservation of bioactivity during the food processing chain, and to provide scientific evidence of the efficacy of key bioactives in foods in preventing disease and improving health and wellbeing.

Abstract

The development and treatment of cancer presents a complex interaction between tumor and host. Provision of nutrients not only enables the maintenance of nutritional status, but also provides substrates and signals for immunity, tumor metabolism and protection of the host from treatment toxicities. Fat is one dietary element that has been explored for its role in cancer development. While the bulk of these studies have been observational or experimental, the evidence assembled suggests that dietary lipids behave uniquely to prevent or promote cancers. An additional aspect of cancer development is the role of adipose tissue as a source of, and a responder to, inflammatory signals that may be involved in tumor development. This Special Issue of Nutrients focuses on fat and cancer. The contributors to this Special Issue are well-recognized leaders in the field of cancer and have unique areas of focus including metabolism, immunology, biochemistry, epidemiology and nutrition. Each contribution highlights the latest research in these areas and what is known about fat and cancer with topics ranging from diet and cancer prevention, mechanisms of n-3 fatty acids on tumor development and the role of adipose tissue in cancer development and progression.

Abstract

Folate, vitamin B12, vitamin B6 and riboflavin play a key role as coenzymes in one-carbon metabolism which, in turn, is essential for a broad range of fundamental physiological processes, including RNA and DNA synthesis, cell division, tissue growth and methylation. Deficiencies or imbalance of B-vitamins, as well as genetic polymorphisms and environmental factors, are shown to disturb the normal function of one-carbon metabolism with adverse effects on human health. Although a vast volume of research has already been conducted in this area, there are still significant gaps in our knowledge that require further investigations. This Special Issue of Nutrients invited submission of manuscripts, original research or reviews of the scientific literature, focused on novel findings in relation to B-vitamins and one-carbon metabolism in terms of: metabolic roles and molecular mechanisms; gene–nutrient interactions; fetal growth and programing; risk of disease (birth defects and pregnancy related conditions, cancer, cardiovascular disease and hypertension, neuropsychiatric disease, osteoporosis); health effects of B-vitamin supplementation and food fortification.

Abstract

The primary function of mitochondria is respiration, where the catabolism of substrates is coupled to ATP synthesis via oxidative phosphorylation. In plants, mitochondrial composition is relatively complex and flexible and has specific pathways to support photosynthetic processes in illuminated leaves. Plant mitochondria also play important roles in a variety of cellular processes associated with carbon, nitrogen, phosphorus, and sulfur metabolism. Research on plant mitochondria has rapidly developed in the last few decades with the availability of the genome sequences for a wide range of model and crop plants. Recent prominent themes in plant mitochondrial research include linking mitochondrial composition to environmental stress responses, and how this oxidative stress impacts on the plant mitochondrial function. Similarly, interest in the signaling capacity of mitochondria, the role of reactive oxygen species, and retrograde and anterograde signaling has revealed the transcriptional changes of stress responsive genes as a framework to define specific signals emanating to and from the mitochondrion. There has also been considerable interest in the unique RNA metabolic processes in plant mitochondria, including RNA transcription, RNA editing, the splicing of group I and group II introns, and RNA degradation and translation. Despite their identification more than 100 years ago, plant mitochondria remain a significant area of research in the plant sciences. This Special Issue, “Plant Mitochondria”, will cover a selection of recent research topics and timely review articles in the field of plant mitochondrial research.

Abstract

Phytoalexins are antimicrobial substances of low molecular weight produced by plants in response to infection or stress, which form part of their active defense mechanisms. It is now clear that phytoalexins exhibit toxicity across much of the biological spectrum, prokaryotic and eukaryotic. Starting in the 1950s, research on phytoalexins has begun with biochemistry and bio-organic chemistry, resulting in the determination of their structure, their biological activity as well as mechanisms of their synthesis and their catabolism by microorganisms. Elucidation of the biosynthesis of numerous phytoalexins has also permitted the use of molecular biology tools for the exploration of the genes encoding enzymes of their synthesis pathways and their regulators. This has led to potential applications for increasing plant resistance to diseases. Phytoalexins display an enormous diversity belonging to various chemical families such as isoflavones, isoflavanones, pterocarpans, isoflavans, flavanones, coumestans, furanoacetylenes, phenylpropanoids, steroid glycoalkaloids, norsesquiterpenoids/sesquiterpenoids, coumarins, diterpenes, ent-kaurane-related diterpenoids, acidic sesquiterpenoids, 3-deoxyanthocyanidins, naphthaldehydes, indoles and stilbenes.Research papers dealing with all aspects of phytoalexins, including structure elucidation; chemical synthesis; methods for phytoalexin analysis in plant extracts or biological fluids; biosynthesis studies including modulation of phytoalexin synthesis; engineering of phytoalexin biochemical pathways in plants and microbes; biological activities; structure/activity relationships; phytoalexin metabolism in planta and by micro-organisms; roles and ATP Binding Cassette (ABC) transporters or Multi-Drugs Efflux (MDE) transporters are welcome for inclusion in this Special Issue of Molecules. Review articles, particularly those dealing with the themes mentioned above are also particularly welcome for inclusion.

Abstract

Currently, we are witnessing highly dynamic research efforts related to the exciting field of novel biodegradable plastic-like materials. These activities originate from a growing public awareness of prevailing ecological problems associated to, e.g., rising piles of plastic waste, increasing greenhouse gas emissions, and ongoing depletion of such fossil resources usually used for the synthesis of “full carbon backbone” plastics. Polyhydroxyalkanoate (PHA) biopolyesters, a family of versatile plastic-like materials produced by living microbes, are a future-oriented alternative to traditional plastics. If accomplished in an optimized way, production and the entire lifecycle of PHA are embedded into nature´s closed carbon cycle, which is underlined by PHA´s main benefits of being “biobased”, “biosynthesized”, “biocompatible”, and “biodegradable”.Sustainable and economically feasible PHA synthesis, especially on an industrially relevant scale, requires all production steps to be understood and improved. Among other aspects, this calls for new powerful production strains to be screened; knowledge about the proteome and genome of PHA accumulating organisms to be consolidated; the kinetics of the bioprocesses to be thoroughly understood; abundantly available inexpensive raw materials to be tested; the monomer composition of PHA to be adapted; (bio)chemical engineering to be optimized; and novel PHA recovery strategies to be developed in order to reduce energy and chemical inventory.The present book provides a comprehensive compilation of articles addressing all these different aspects; the individual chapters were composed by globally recognized front running experts from special niches of PHA research. We are convinced that this book will be of major benefit to the growing scientific community active in biopolymer research.
