Abstract

In the genomic era of 1990s-2000s, pharmaceutical research moved to target-based drug discovery which enabled development of a number of small molecule drugs against a wide range of diseases. In many cases however, drugs that arose from genomics failed, questioning the validity of the targets and the suitability of target-based drug discovery as an optimal strategy for all disease states. For monogenic diseases, target-based approaches may be well-suited to the identification of novel therapies. Most diseases, however, are caused by a combination of several genetic and environmental factors and are likely to require simultaneous modulation of multiple molecular targets/pathways for successful treatment. For such diseases, reductionist approaches focusing on individual targets rather than biological networks are unlikely to succeed and new drug development strategies are required. In search of more successful approaches, the pharmaceutical industry is moving towards phenotypic screening beyond individual genes/targets. However, this requires rethinking of diseases and drug discovery approaches from a network and systems biology perspective. Since returning to the pre-genomics era of screening drug candidates in laborious animal models is not a feasible solution, the industry needs to evolve a new paradigm of phenotypic drug discovery within the context of systems biology. Such a paradigm must combine physiologically and disease relevant biological substrates with sufficient throughput, operational simplicity and statistical vigour. Biomarker strategies for translational medicine, as well as preclinical safety and selectivity assessments, would also need to be revised to adapt to the target agnostic style. This focused issue aims to discuss strategies, key concepts and technologies related to systems-based approaches in drug development. Design and implementation of innovative biological assays, featuring multiple target strategies, and rational drug design in the absence of target knowledge during the early drug discovery are illustrated with examples. Specific topics include: • The need for systems-based approaches in drug development • Phenotypic screening strategies • Compound libraries (natural product inspired compound collections) • Target deconvolution and identification • Target agnostic lead discovery and optimization • Multi-target approaches and decoding the phenotype (understanding biological interactions and multiscale systems modelling) • Translational aspects • Early evaluation of selectivity and safety in a target agnostic manner

Abstract

Abstract

In Western countries, cancer is among the most frequent causes of death. Despite striking advances in cancer therapy, especially by the so called “targeted agents”, there is still an urgent need for new drugs in oncology. Hope comes from the marine environment, which is a rich source of natural compounds showing anti-cancer activity. To date, four marine cytotoxic substances, namely cytarabine, trabectidine, eribulin, and monomethyl auristatin E (as a drug-antibody conjugate named vedotin) have made it into clinical routine. Many more are in all phases of clinical testing, and a plethora of substances has already been examined for in vitro and in vivo activity.Interestingly, more and more precise research tools allow the dissection of the molecular mode of action of these cytotoxic substances, thereby uncovering the specific drug targets in cancer cells. This development will blur the edges between “targeted” and “untargeted” therapy, and will hopefully lead to a more directed use of cancer medicine (based on a molecular rationale of activity) in the future.This Topical Collection will cover the whole scope from agents with cancer-preventive activity, to novel and previously characterized compounds with anti-cancer activity, both in vitro and in vivo, and the latest status of clinical development from drug trials. Of note, compounds possessing pro-carcinogenic activity or mediating cancer cell survival are also within the scope of this Topical Collection. In addition, a special focus will be placed on current shortfalls and possible strategies to overcome obstacles in the area of marine anti-cancer drug development.

Abstract

The importance of bioactive natural compounds in pharmacology and other biotechnological fields has stimulated the scientific community to explore new environmental contexts and their associated microbial diversity. As the largest frontier in biological discovery, the sea represents a significant source of organisms producing novel secondary metabolites with interesting bioactivities. Of the available biological material, fungi have received increasing consideration, both due to their pervasive occurrence in varying habitats as well as their aptitude to develop symbiotic associations with higher organisms in numerous contexts. In many cases, fungal strains have been reported as the real producers of drugs originally extracted from marine plants and animals. Due to the constantly increasing number of marine-derived fungi yielding valuable bioactive products, it is now appropriate to present these findings to a recipient audience in a more organized form. This Special Issue of Marine Drugs, entitled “Bioactive Compounds from Marine-Derived Aspergillus, Penicillium, Talaromyces, and Trichoderma Species"" is specifically focused on a few genera of ascomycetous fungi which are widespread regarding marine contexts and are particularly inclined to establishing symbiotic relationships. For this project, we welcome submissions of full research papers, short notes, and review articles reporting the discovery and characterization of products showing antibiotic, antitumor, antiviral, insecticidal, antimalarial, antifouling, antioxidant, plant growth-promoting and/or resistance-inducing, as well as other less-exploited activities.
