Abstract

Neuroanatomists increasingly rely on techniques enabling them to manipulate genes in defined brain cell populations. In particular, engineered transgenes, which encode a variety of fluorescent reporter proteins can be inserted into the genome or delivered into desired brain regions using viral vectors, thereby allowing the labeling of molecularly-defined populations of neurons and/or glial cells. Transgenic technology can also be used to selectively delete genes in targeted neuronal populations or bi-directionally modulate their electrical excitability using optogenetic or chemogenetic techniques. One of the primary advantages of using transgenic reagents is to simplify the identification and tracing of targeted population of brain cells, which can be laborious using traditional techniques in neuroanatomy. In this research topic, we assembled up-to-date reviews and original articles that demonstrate the versatility and power of transgenic tools in advancing our knowledge of the nervous system, with a special emphasis on the application of transgenic technology to neuroanatomical questions.

Abstract

Understanding the underlying mechanisms of how axons and dendrites develop is a fundamental problem in neuroscience and a main goal of research on nervous system development and regeneration. Previous studies have provided a tremendous amount of information on signaling and cytoskeletal proteins regulating axonal and dendritic growth and guidance. However, relatively little is known about the relative contribution and role of cytoskeletal dynamics, transport of organelles and cytoskeletal components, and force generation to axonal elongation. Advancing the knowledge of these biomechanical processes is critical to better understand the development of the nervous system, the pathological progression of neurodegenerative diseases, acute traumatic injury, and for designing novel approaches to promote neuronal regeneration following disease, stroke, or trauma. Mechanical properties and forces shape the development of the nervous system from the cellular up to the organ level. Recent advances in quantitative live cell imaging, biophysical, and nanotechnological methods such as traction force microscopy, optical tweezers, and atomic force microscopy have enabled researchers to gain better insights into how cytoskeletal dynamics and motor-driven transport, membrane-dynamics, adhesion, and substrate rigidity influence axonal elongation. Given the complexity and mechanical nature of this problem, mathematical modeling contributes significantly to our understanding of neuronal mechanics. Nonetheless, there has been limited direct interaction and discussions between experimentalists and theoreticians in this research area. The purpose of this Frontiers Research Topic is to highlight exciting and important work that is currently developing in the fields of neuronal cell biology, neuronal mechanics, intracellular transport, and mathematical modeling in the form of primary research articles, reviews, perspectives, and commentaries.

Abstract

Cerebral cortex is probably the most complex biological network. Here many millions of individual neurons, the functional units of cortex, are interconnected through a massive yet highly organized pattern of axonal and dendritic wiring. This wiring enables both near and distant cells to coordinate their responses and generate a rich variety of cognitions and behaviours. When the wiring is damaged through disease or trauma it may reorganize but this may lead to characteristic pathological behaviours. While there have been significant advances in mapping cortical connectivity, the organizing principles and function of this connectivity are not well understood. On the one hand, there appears to be general design constraints governing cortical wiring, as first recognised by Rámon y Cajal's in his laws of conduction, material, and volume conservation. Yet on the other hand, particular patterns of cortical wiring exist to serve specific functions. There is a wide gap in understanding how the response and connectivity properties of a single neuron contribute to emergent network functions such as in detecting perceptually relevant features. Unravelling this intimate causal relationship represents one of the major challenges in neuroscience. This Research Topic will examine progress in understanding cortical wiring principles. This Research Topic aims to draw together recent advances in methods and understanding as well as recent challenges to existing ideas about how cerebral cortex is wired. This is particularly timely because new automated techniques may soon yield huge datasets in need of explanation. Recent studies have, for instance, empirically evaluated Rámon y Cajal's conservation laws for cerebral cortex, while others have shown some unexpected connectivity features that may refine the traditional view of how corticocortical connections are organised with regard to functional representations of auditory, somatosensory and visual cortices. Understanding these data will help improve the fidelity of neural models of cerebral cortical function and take into account the diversity of connections at both micro- and mesoscopic scales not seen at such a depth before.

Abstract

"This grammar provides a grammatical description of Palula, an Indo-Aryan language of the Shina group. The language is spoken by about 10,000 people in the Chitral district in Pakistan’s Khyber Pakhtunkhwa Province. This is the first extensive description of the formerly little-documented Palula language, and is one of only a few in-depth studies available for languages in the extremely multilingual Hindukush-Karakoram region. The grammar is based on original fieldwork data, collected over the course of about ten years, commencing in 1998. It is primarily in the form of recorded, mainly narrative, texts, but supplemented by targeted elicitation as well as notes of observed language use. All fieldwork was conducted in close collaboration with the Palula-speaking community, and a number of native speakers took active part in the process of data gathering, annotation and data management. The main areas covered are phonology, morphology and syntax, illustrated with a large number of example items and utterances, but also a few selected lexical topics of some prominence have received a more detailed treatment as part of the morphosyntactic structure. Suggestions for further research that should be undertaken are given throughout the grammar. The approach is theory-informed rather than theory-driven, but an underlying functional-typological framework is assumed. Diachronic development is taken into account, particularly in the area of morphology, and comparisons with other languages and references to areal phenomena are included insofar as they are motivated and available. The description also provides a brief introduction to the speaker community and their immediate environment. "

Abstract

This book presents a synchronic grammar of the southern dialects of Yauyos, an extremely endangered Quechuan language spoken in the Peruvian Andes. As the language is highly synthetic, the grammar focuses principally on morphology; a longer section is dedicated to the language's unusual evidential system. The grammar's 1400 examples are drawn from a 24-hour corpus of transcribed recordings collected in the course of the documentation of the language.

Abstract

Recent years have seen an upsurge of interest in the notion of salience in linguistics and related disciplines. While in top-down salience, perceivers endogenously direct their attention to a certain stimulus, in the bottom-up salience, it is the stimulus itself which attracts attention. In prototypical cases of bottom-up salience, the stimulus stands out because it is incongruous with a given ground by virtue of intrinsic physical characteristics. But a stimulus may also cause surprise by virtue of deviating from a cognitive ground, e.g., when violating social or probabilistic expectations. This has prompted researchers to examine the relationship between expectations and the perceptual salience of linguistic stimuli in new ways. This e-book features contributions from different scientific frameworks. The reader will find commentaries, reviews, and original research articles on models of sociolinguistic and morphological salience, the role of attention, affect, and predictability, and on how salient items are processed, categorized and learned. Taken together, the articles in this volume contribute to our understanding of how the perceptual salience of linguistic forms and variants can be theoretically framed and methodologically operationalized in different areas of linguistic processing.

Abstract

In June 2015 we held a workshop on the beautiful island of Mallorca, Spain with a focus on sea level variability and change. Over 120 sea level experts from around the world attended this workshop, from a range of different disciplines. The main aims of the workshop were to: 1.) Evaluate the current state-of-knowledge of sea level science; 2.) Identify gaps and unresolved questions in any aspect of sea level science; and 3.) Design future research to address these issue. All aspects of sea level changes were covered, from global to regional, observations and modelling, processes driving mean sea level changes and extremes, from the geological scale to the instrumental era and future projections and including impacts on the coastal zones. This E-Book presents papers that came out of that workshop. Overall, these papers illustrate the multi-disciplinary nature of sea level research, cross-cutting many fields of research including: oceanography, meteorology, geology, coastal morphodynamics, engineering and the social-economic aspects. Collectively, theses articles represent an interesting range of perspectives and original studies that contribute to understanding the dynamic nature of sea level and its impacts across a wide range of time and space scales. Abstract

This collection attempts to integrate work pertaining to a fundamental question in plant evolution: What are the molecular underpinnings for the origin of different plant forms? Among the many facets this question touches are the transition to land, the emergence of vascular plants, the origin of the seed and the origin and diversification of floral form. We aim to bring to the forefront the most salient and original plant systems and approaches within an inclusive phylogenetic context that encompasses representatives of the major lineages of land plants.
