Abstract

Mitogen-activated protein kinase (MAPK) pathways are evolutionarily conserved in all eukaryotes and allow cells to respond to changes in the physical and chemical properties of the environment and to produce an appropriate response by altering many cellular functions. MAPKs are among the most intensively studied signal transduction systems. MAPK research is a very dynamic field in which new perspectives are continuously opening to the scientific community. Importantly, many MAPK inhibitors have been developed during the last years and are currently being tested in preclinical and clinical assays for inflammatory diseases and cancer treatment. In this research topic, we have gathered 14 papers covering recent advances in different aspects of the MAPK research area that have provided valuable insight into the spatiotemporal dynamics, the regulation and functions of MAPK pathways, as well as their therapeutic potential. We hope that this Research Topic helps readers to have a better understanding of the progresses that have been made recently in the field of MAPK signalling. A deeper understanding of the these pathways will facilitate the development of innovative therapeutic approaches.

Abstract

The adipokine adiponectin is very concentrated in plasma, and decreased levels of adiponectin are associated with pathological conditions such as obesity, diabetes, cardiovascular diseases, and metabolic syndrome. When produced in its full-length form, adiponectin self-associates to generate multimeric complexes. The full-length form of adiponectin can be cleaved by the globular form of elastase that is produced locally, and the resulting biological effects are exerted in a paracrine or autocrine manner. The different forms of adiponectin bind to specific receptors consisting of two G-protein-independent, seven-transmembrane-spanning receptors, called AdipoR1 and AdipoR2, while T-cadherin has been identified as a potential receptor for high molecular weight complexes of adiponectin. Adiponectin exerts a key role in cellular metabolism, regulating glucose levels as well as fatty acid breakdown. However, its biological effects are heterogeneous, involving multiple target tissues. The Special Issue “Mechanisms of Adiponectin Action” highlights the pleiotropic role of this hormone through 3 research articles and 7 reviews. These papers focus on the recent knowledge regarding adiponectin in different target tissues, both in healthy and in diseased conditions.

Abstract

Glycans (carbohydrate chains) of marine creatures are rich and diverse in polysaccharides, glycoproteins, and glycolipids. The chains that are metabolized by glycan-related enzymes (glycosyltransferases and glycosidases) are recognized by glycan-binding proteins (lectins) which regulate cellular processes such as growth, differentiation, and death. Marine glycomics that involves the genome and transcriptome accelerates our understanding of the evolution of glycans, glycan-related enzymes, and lectins. From 2017 to 2019, the Special Issue “Marine Glycobiology, Glycomics and Lectins” of the journal Marine Drugs published scientific articles and reviews, on the background of “glycobiology”—that is, glycan-based biosciences. The aim was to promote the discovery of novel biomolecules that contribute to drug development and clinical studies. This has great potential for establishing connections between the fields of both human health and marine life sciences.This book contains 11 scientific papers representing current topics in comprehensive glycosciences related to therapeutic agents from marine natural products, as outlined.
