Abstract

The 8th International Conference on Protein Kinase CK2 (www.uks.eu/ck2), organized by Matthias Montenarh and Claudia Götz, will be held at the Faculty of Medicine of Saarland University, in Homburg, Germany, from 6–9 September, 2016.Protein kinase CK2 is a serine/threonine kinase, which is highly conserved in evolution. Knock-out experiments have shown that this enzyme is absolutely necessary for cell viability. The enzyme is highly expressed in rapidly proliferating cells, which goes along with an elevated enzyme activity. Protein kinase CK2 is involved in a broad range of signaling pathways, regulating proliferation, differentiation, apoptosis, or senescence. Recent advances towards the characterization of the three-dimensional structure of protein kinase CK2 and its subunits will undoubtedly yield important new insights into its regulation, and the functions of CK2. Moreover, with two inhibitors of the enzyme presently in clinical phase II trials, human protein kinase CK2 has appeared as an emerging target for cancer diseases.

Abstract

The peripheral membrane proteins transduce the extracellular signaling into the cells. The peripheral membrane proteins include the G protein-coupled receptors, receptor tyrosine kinases, channels and transporters. The signals, which are transduced via the peripheral membrane proteins, activate several pathways including G protein signaling, MAPK signaling, TNF signaling, TGF? signaling, Wnt signaling and Hedgehog signaling. The peripheral membrane proteins transduce the signaling from the extracellular ligands into the cells. Abstract

DNA damage response (DDR) and lesion repair are vital processes ensuring genome integrity through various pathways depending mainly on the nature of DNA injury and cell cycle stage. DDR is finely regulated at many levels in co-ordination with other ongoing processes as is genome replication and cell cycle progression. Posttranslational modifications (PTMs), affecting both protein-protein and protein-DNA interactions, play a crucial role in finely tuning all processes involved in the restoration of genome lesions. Regarding damaged chromatin, PTMs serve in many cases as recruitment platforms for DNA repair mechanisms by facilitating binding sites or regulating interactions between involved proteins. Ubiquitination, the addition of ubiquitin moieties on a target protein, apart from controlling protein availability through degradation, is also involved, together with partner small ubiquitin-like modifier (SUMO), in controlling many pathways involved in DDR by modifying the structure-function relationship and thus interacting with partner molecules. The aim of this book is to cover a broad spectrum of current topics in ubiquitination and to a lesser extent SUMOylation involvement in regulation of DDR and repair in health and disease. This book is intended for pre- and postgraduate students and young scientists in this field. Members of both academic and research institutions, actively involved in the field, have described their current understanding of major mechanisms involved, highlighted key events, described ongoing applications in both developmental diseases and cancer and provided hints for future potential applications.

Abstract

The human ubiquitin proteasome system (UPS) is comprised of nearly 1000 proteins. Although originally identified as a mechanism of protein destruction, the UPS has numerous additional functions and mediates central signaling events in myriad processes involved in both cellular and organismal health and homeostasis. Numerous pathways within the UPS are implicated in disease, ranging from cancer to neurodegenerative diseases such as Parkinson's. The goal of this book is to deliver a collection of synopses of current areas of UPS research that highlights the importance of understanding the biology of the UPS to identify disease-relevant pathways, and the need to elucidate the molecular machinations within the UPS to develop methods for therapeutic modulation of these pathways.

Abstract

Interest in understanding the biological role of carbohydrates has increased significantly over the last 20 years. The use of structural techniques to understand carbohydrate-protein recognition is still a relatively young area, but one that is of emerging importance. The high flexibility of carbohydrates significantly complicates the determination of high quality structures of their complexes with proteins. Specialized techniques are often required to understand the complexity of carbohydrate recognition by proteins. In this Research Topic, we will focus on structural and computational approaches to understanding carbohydrate recognition by proteins involved in immunity and infection. Particular areas of focus include cancer immunotherapeutics, carbohydrate-lectin interactions, glycosylation and glycosyltransferases.

Abstract

Viruses are obligate intracellular parasites that need to co-opt a living cell’s machinery for replication. At the heart of the viral replication machinery are the nucleic acid polymerases, which are responsible for efficiently copying the viral genome. This process must often be coordinated with other viral processes including protein translation and viral packaging. The nucleic acid polymerases may also be responsible for generating genetic diversity that is important for escape from the host’s defenses. The polymerases and other components of the replication machinery may serve as potential anti-viral targets. In this Special Issue, we seek to highlight recent advances into uncovering the structure and function of viral replication complexes. We are especially seeking to highlight the wide-range of methodologies used to gain structural insight into viral replication. Topics of interest include molecular mechanisms of nucleic acid polymerases, nucleic acid recognition and translocation, fidelity and error correction, interactions with lipid membranes, and coordination/regulation of transcription, translation and replication processes.

Abstract

Viruses in the Parvoviridae family constitute one of the most diverse and intriguing fields of research. While they all share an ssDNA genome and a small capsid, they can differ widely in structure, genome organization and expression, virus–cell interaction, and impact on the host. Exploring such diversity and unraveling the inherent complexity in these apparently simple viruses is an ongoing endeavor and commitment for the scientific community. The translational implications of research on parvoviruses are relevant. Within the family, some viruses are important human and veterinary pathogens, in need of diagnostic methods and antiviral strategies; other viruses have long been studied and engineered as tools for oncolytic therapy, or as sophisticated gene delivery vectors, and can now display their wide and expanding applicative potential. This Special Issue of Viruses collects recent contributions in the field of parvovirus research, with a focus on new insights and research on unresolved issues, as well as new approaches exploiting systemic methodologies. Evolution, structural biology, viral replication, virus–host interaction, pathogenesis and immunity, and viral oncotherapy are a selection of the topics addressed in the issue that can be of relevance to the community involved in parvovirus research and of interest to a wider audience.
