Interaction Between Hyaluronic Acid and Its Receptors (CD44, RHAMM) Regulates the Activity of Inflammation and Cancer

Abstract

The biological outcome of Hyaluronan (also hyaluronic acid, abbreviated HA) interaction with its CD44 or RHAMM receptors recently attracted much attention within the scientific community owing to a Nature article by Tian X et al. (Nature 2013; 499:346-9). The article described a life span exceeding 30 years in naked mole rats, whereas the maximal lifespan of mice, to which the naked mole rat is related, is only 4 years. This observation is accompanied by the finding that the naked mole rat, in contrast to the mouse, does not develop spontaneous tumors during this exceptional longevity. The article provides evidence that interaction of long tissue HA (6000-12,000 kDa) of the naked mole rat with cell surface CD44, in contrast to the interaction of short tissue HA (less than 3000 kDa) with the mouse CD44, makes the difference. More specifically, this communication shows that the interaction of short HA with fibroblasts’ CD44 imposes on them susceptibility for malignant transformation, whereas the corresponding interaction with long HA imposes on the fibroblasts a resistance to malignant transformation. The article does not explain the mechanism that underlines these findings. However, the articles, that will be published in the proposed Research Topic in the Inflammation section of Frontiers in Immunology, can bridge not only this gap, but also may explain why interaction between short HA and cell surface CD44 (or RHAMM, an additional HA receptor) enhances the development of inflammatory and malignant diseases. Furthermore, the articles included in the proposed Frontiers Research Topic will show that cancer cells and inflammatory cells share several properties related to the interaction between short HA and cell surface CD44 and/or RHAMM. These shared properties include: 1. Support of cell migration, which allows tumor metastasis and accumulation of inflammatory cells at the inflammation site; 2. Delivery of intracellular signaling, which leads to cell survival of either cancer cells or inflammatory cells; 3. Delivery of intracellular signaling, which activates cell replication and population expansion of either cancer cells or inflammatory cells; and 4. Binding of growth factors to cell surface CD44 of cancer cells or inflammatory cells (i.e., the growth factors) and their presentation to cells with cognate receptors (endothelial cells, fibroblasts), leading to pro-malignant or pro-inflammatory activities. Going back to the naked mole rat story, we may conclude from the proposed articles of this Frontiers Research Topic that the long HA, which displays anti-malignant effect, interferes with the above described pro-malignant potential of the short HA (perhaps by competing on the same CD44 receptor). Extrapolating this concept to Inflammation, the same mechanism (competition?) may be valid for inflammatory (and autoimmune) activities. If this is the case, long HA may be used for therapy of both malignant and inflammatory diseases. Moreover, targeting the interaction between short HA and CD44 (e.g. by anti-CD44 blocking antibodies) may display also a therapeutic effect on both malignant and inflammatory diseases, an issue that encourages not only fruitful exchange of views, but also practical experimental collaboration.

Abstract

Through evolution of life, animals have adapted to the ubiquitous presence of metals in the biosphere. They utilize the more frequent ones as essential constituents of their biochemical machinery. In fact, about 40% of all proteins present in animal cells are so-called metalloproteins. On the other hand, animals have invented regulatory and detoxifying mechanisms to protect themselves from critical concentrations of both essential and non-essential metal concentrations. Metallomics is a modern approach applying cellular, biochemical, molecular and analytical methods to investigate the relationships of metals in their cellular context. The present edition contains a number of original articles and reviews dealing with various aspects of metallomics in animals, published as Special Issues of the International Journal of Molecular Sciences in 2016 and 2017. The book addresses subjects such as metal definition in biology, metabolism of metals in invertebrate and vertebrate animals, metal detoxification and regulation strategies, supplementation of essential trace elements, metal behavior in pregnancy and embryonic development, as well as metal toxicology and emerging medical implications.

Abstract

Aging of unicellular and multicellular eukaryotic organisms is a convoluted biological phenomenon, which is manifested as an age-related functional decline caused by progressive dysregulation of certain cellular and organismal processes. Many chronic diseases are associated with human aging. These aging-associated diseases include cardiovascular diseases, chronic obstructive pulmonary disease, chronic kidney disease, diabetes, osteoarthritis, osteoporosis, sarcopenia, stroke, neurodegenerative diseases (including Parkinson’s, Alzheimer’s, and Huntington’s diseases), and many forms of cancer. Studies in yeast, roundworms, fruit flies, fishes, mice, primates, and humans have provided evidence that the major aspects and basic mechanisms of aging and aging-associated pathology are conserved across phyla. The focus of this International Journal of Molecular Sciences Special Issue is on molecular and cellular mechanisms, diagnostics, and therapies and diseases of aging. Fifteen original research and review articles in this Special Issue provide important insights into how various genetic, dietary, and pharmacological interventions can affect certain longevity-defining cellular and organismal processes to delay aging and postpone the onset of age-related pathologies in evolutionarily diverse organisms. These articles outline the most important unanswered questions and directions for future research in the vibrant and rapidly evolving fields of mechanisms of biological aging, aging-associated diseases, and aging-delaying therapies.

Abstract

Two review papers, eight research articles, and one brief report were published in this Special Issue. They showed the rich resources that are present within the genomes of marine microorganisms and discussed the use of recently developed tools and technologies to exploit this genetic richness. Examples include the rational supply of precursors according to the relevant biosynthetic pathway and stress driven discovery together with the use of histone deacetylase inhibitors to facilitate the discovery of new bioactive molecules with potential biopharmaceutical applications. We believe that the content of this Special Issue reflects the current state-of-the-art research in this area and highlights the interesting strategies that are being employed to uncover increasing numbers of exciting novel compounds for drug discovery from marine genetic resources.

Abstract

Sterols and other isoprenoids are of great interest for their molecular structure and function in cell architecture and evolution, as well as for their importance in medicine and agriculture. Molecules’ 2019 Festschrift Special Issue in honor of the 65th birthday of Prof. W. David Nes, an internationally recognized chemical biologist and recipient of the George Schroepher medal for sterol research, focuses on recent developments in the chemistry, biosynthesis, and function of these polycyclic natural products. This volume of Molecules contains 16 leading-edge review articles and original research contributions from an international cast of scientists. This volume is grouped into three sections: (i) isoprenoid metabolome and diversity, (ii) clinical evaluation of sterol and triterpene structures and biosynthesis, and (iii) methods and synthesis of steroids and other compounds. The volume will be a valuable reference tool for those who study medicinal chemistry, protein chemistry, and biochemistry of isoprenoid lipids.

Abstract

Plants possess a rather complex and efficient immune system. During their evolutionary history, plants have developed various defense strategies in order to recognize and distinguishing between self and non-self, and face pathogens and animal pests. Accordingly, to study the plant innate immunity represents a new frontier in the plant pathology and crop protection fields. This book is structured in 6 sections. The first part introduces some basic and general aspects of the plant innate immunity and crop protection. Sections 2–5 focus on fungal and oomycete diseases (section 2), bacterial and phytoplasma diseases (section 3), virus diseases (section 4), and insect pests (section 5), with a number of case studies and plant–pathogen/pest interactions. The last section deals with plant disease detection and control. The book aims to highlight new trends in these relevant areas of plant sciences, providing a global perspective that is useful for future and innovative ideas.

Abstract

Antioxidative polyphenols represented by tannins and flavonoids are rich in numerous food sources and traditional natural medicines and currently attracting increased attention in health care and food industries because of their multiple biological activities that are favorable to human health. Commemorating the outstanding achievements on tannins by Dr. Takuo Okuda on the occasion of his passing away in December 2016, his colleagues, friends, and worldwide experts of polyphenol research have contributed 18 papers on their recent study to the Special Issue of Molecules. This book is its reprinted form. This covers reviews of structural features, historical usages, and biological activities of unique class of ellagitannins and condensed tannins, and original articles on the most up-to-date findings on the anticancer effect of green tea catechins, the antivirus effect of tannins comparing with the clinically used drugs, the analytical method of ellagitannins using quantitative NMR, the chemical structures of Hydrangea-blue complex (pigment) and condensed tannins in Ephedra sinica and purple prairie clover, and the relationship of condensed tannins in legumes and grape-marc with methane production in the in vitro ruminant system, and others. This book will be useful to natural product chemists and also to researchers in pharmaceutical and/or food industry.

Abstract

Polyamines are ubiquitous polycations essential for all cellular life. The most common polyamines in eukaryotes, spermine, spermidine, and putrescine, exist in millimolar intracellular concentrations that are tightly regulated through biosynthesis, catabolism, and transport. Polyamines interact with, and regulate, negatively charged macromolecules, including nucleic acids, proteins, and ion channels. Accordingly, alterations in polyamine metabolism affect cellular proliferation and survival through changes in gene expression and transcription, translation, autophagy, oxidative stress, and apoptosis. Dysregulation of these multifaceted polyamine functions contribute to multiple disease processes, thus their metabolism and function have been targeted for preventive or therapeutic intervention. The correlation between elevated polyamine levels and cancer is well established, and ornithine decarboxylase, the rate-limiting biosynthetic enzyme in the production of putrescine, is a bona fide transcriptional target of the Myc oncogene. Furthermore, induced polyamine catabolism contributes to carcinogenesis that is associated with certain forms of chronic infection and/or inflammation through the production of reactive oxygen species. These and other characteristics specific to cancer cells have led to the development of polyamine-based agents and inhibitors aimed at exploiting the polyamine metabolic pathway for chemotherapeutic and chemopreventive benefit. In addition to cancer, polyamines are involved in the pathologies of neurodegenerative diseases including Alzheimer’s and Parkinson’s, parasitic and infectious diseases, wound healing, ischemia/reperfusion injuries, and certain age-related conditions, as polyamines are known to decrease with age. As in cancer, polyamine-based therapies for these conditions are an area of active investigation. With recent advances in immunotherapy, interest has increased regarding polyamine-associated modulation of immune responses, as well as potential immunoregulation of polyamine metabolism, the results of which could have relevance to multiple disease processes. The goal of this Special Issue of Medical Sciences is to present the most recent advances in polyamine research as it relates to health, disease, and/or therapy.
