Abstract

Soft tissue and bone sarcomas disproportionately affect children, adolescents, and young adults. Although considered a rare disease, sarcomas continue to have a devastating effect on these patients and their loved ones, and their impact on our society far exceed their relatively low prevalence. Because of its rarity and heterogeneity, clinical decision making on management of sarcomas are often individualized and without a consensus treatment strategy. Furthermore, research to improve clinical outcomes are particularly challenging as sizeable studies are hard to produce. Abstract

Traumatic brain injury (TBI) is a major cause of death and disability and one of the greatest unmet needs in medicine and public health. TBI not only has devastating effects on patients and their relatives but results in huge direct and indirect costs to society. Although guidelines for the management of patients have been developed and more than 200 clinical trials have been conducted, they have resulted in few improvements in clinical outcomes and no effective therapies approved for TBI. It is now apparent that the heterogeneity of clinical TBI is underlain by molecular phenotypes more complex and interactive than initially conceived and current approaches to the characterization, management and outcome prediction of TBI are antiquated, unidimensional and inadequate to capture the interindividual pathophysiological heterogeneity. Recent advances in proteomics and biomarker development provide unparalleled opportunities for unraveling substantial injury-specific and patient-specific variability and refining disease characterization. The identification of novel, sensitive, objective tools, referred to as biomarkers, can revolutionize pathophysiological insights, enable targeted therapies and personalized approaches to clinical management. In this Research Topic, we present novel approaches that provide an infrastructure for discovery and validation of new biomarkers of acute brain injury. These techniques include refined mass spectrometry technology and high throughput immunoblot techniques. Output from these approaches can identify potential candidate biomarkers employing systems biology and data mining methods. In this Research Topic, we present novel approaches that provide an infrastructure for discovery and validation of new biomarkers of acute brain injury. Abstract

Our understanding of the mechanisms underlying the development of multifactorial diseases such as diabetes, autism, Alzheimer's disease, and cancer has been greatly advanced. Non-coding RNAs (ncRNAs), generally including microRNAs and long non-coding RNAs, have recently been found to have potential roles in these diseases, and provide new opportunities for developing both specific biomarkers and therapeutic targets. However, the molecular function and regulation of these RNAs still remains challenging. Numerous studies are focusing on this field in order to fully appreciate the role and regulation of these molecules in human medicine and biology. This e-book aims to bring together new findings on Non-coding RNAs in different complex diseases. It will highlight the characterization, roles, mechanism, and mode of action of these RNAs in complex disorders. We believe that the publications on this topic would be exponentially extended in future. The improved approaches at multiple levels may pave the way for designing and applying new biomarker and therapeutic targets for specific diseases based on these attractive molecules.

Abstract

Increasing age has been associated with an insufficient protection following vaccination and an increased incidence and severity of infectious diseases. The predicted acceleration of global population aging will accentuate the need to understand the mechanisms that drive the age-related decline of the immune system and to, eventually, identify strategies to lower the burden of infectious diseases in elderly people. One type of immune cell appears to be most dramatically affected by the aging process: T lymphocytes. Age-related changes of the bone marrow and the thymus microenvironment lead to a decreased thymic output of functional, naïve T lymphocytes. As T lymphocytes are key players of the adaptive immune system, this research topic will summarize our current understanding on how aging affects the microenvironmental niches and molecular networks that are important for the generation, survival and function of naïve, memory and effector T lymphocytes. This research topic will also address the impact of aging on the different T lymphocyte lineages, such as regulatory T cells and Th17 cells and how age-related changes of the microenvironment affect organ- and tissue-resident memory T lymphocytes. Eventually, the identification of a set of markers for immunosenescence would facilitate the design and application of more specific therapies and improved vaccines and vaccination strategies for elderly people, thereby increasing life and health span.

Abstract

In the last decade, sleep spindles have attracted steadily increasing attention. This interest is motivated by the many intriguing relationships between spindles and various diseases (e.g., schizophrenia, Parkinson, Alzheimer, autism, mental retardation), recovery processes (e.g., post brain stroke), and cognitive faculties (e.g., memory consolidation, intelligence, dream recall, sleep preservation). Nonetheless, a methodological wall has impeded the study of sleep spindles. Their investigation rests heavily on our ability to reliably and consistently identify spindle patterns from background EEG activity, a task involving many obstacles, including: a fuzzy definition of spindles, low inter-expert agreement on their scoring, lack of consensus on standard techniques for their automated detection, low reproducibility of observed characteristics and correlates, unavailability of large, standardized, high-quality databases, and inconsistencies in the methods used to evaluate the performance of automated detectors. The primary aims of this research topic were to bring together world-class researchers on a project designed to facilitate exchanges on methodological difficulties encountered in assessing sleep spindles and to promote standardized spindle-related resources. In preparing their contributions, authors were encouraged to use existing – or to propose new – publicly available resources for assessing sleep spindles. To allow fair and accurate comparison of reported results, the authors were also encouraged to validate their tools on a common benchmark. A database containing expert spindle scoring (i.e., the Montreal Archive of Sleep Studies) was made publicly available for that purpose.

Abstract

MicroRNAs (miRNAs) constitute a large family of small, approximately 20–22-nucleotide, non-coding RNAs that are involved in gene regulation, mainly at the post-transcriptional level. Multiple lines of evidence have indicated that miRNAs play important roles in the maintenance of biological homeostasis and that aberrant expression levels of miRNAs are involved in the onset of many diseases, including cancer. In various types of cancer, miRNAs play important roles in tumor initiation and development. Recently, miRNAs have been demonstrated to also be secreted via small endosome-derived vesicles called exosomes—which are derived from multiple cell types—including immunocytes and cancer cells. Exosomal miRNAs exert important functions in cell-to-cell communication and have been investigated as prognostic and diagnostic biomarkers.

Abstract

The prevalence of neurodegenerative disorders is increasing dramatically and one of the major challenges today is the need of early and accurate diagnosis, the other is the need of more effective therapies -in turn the development of such therapies also requires early and accurate diagnosis-. The main hope for an earlier and more accurate diagnosis comes from the use of biomarkers. Much research is being done trying to solve the many interrogates related to the role of biomarkers in clinical practice, including the early diagnosis, differential diagnosis and follow-up of neurodegenerative disorders. This is a field where translational research is intense enough to make this topic interesting for basic researchers and clinicians. Indeed, the amount and quality of articles received in response to the call for contributions was very good. This eBook contains a good amount of high quality articles devoted to diverse techniques across several neurodegenerative disorders from different perspectives, including original reports, reviews, methods reports and opinion letters on biochemical biomarkers in biological fluids, neuroimaging techniques and multidimensional approaches linking clinical findings with biomarkers. The disorders covered are also diverse: Alzheimer’s disease, Frontotemporal Dementia, Dementia with Lewy Bodies, Huntington’s disease, Parkinson’s disease among others. As we can learn from articles in this Research Topic, biomarkers are allowing us to expand the knowledge on the biological and anatomical basis of neurodegenerative diseases and to implement diagnostic techniques in clinical practice and clinical trials.

Abstract

Classically, anti-cancer therapies have always been applied with the primary aim of tumor debulking achieved through widespread induction of cancer cell death. While the role of host immune system is frequently considered as host protective in various (antigen-bearing) pathologies or infections yet in case of cancer overtime it was proposed that the host immune system either plays no role in therapeutic efficacy or plays a limited role that is therapeutically unemployable. The concept that the immune system is dispensable for the efficacy of anticancer therapies lingered on for a substantial amount of time; not only because evidence supporting the claim that anti-cancer immunity played a role were mainly contradictory, but also largely because it was considered acceptable (and sometimes still is) to test anticancer therapies in immunodeficient mice (i.e. SCID/athymic mice lacking adaptive immune system). This latter practice played a detrimental role in appreciating the role of anticancer immunity in cancer therapy. This scenario is epitomized by the fact that for a long time the very existence of cancer-associated antigens or cancer-associated ‘danger signaling’ remained controversial. However, over last several years this dogmatic view has been considerably modified. The existence of cancer-associated antigens and ‘danger signaling’ has been proven to be incontrovertible. These developments have together paved way for the establishment of the attractive concept of “immunogenic cell death” (ICD). It has been established that a restricted class of chemotherapeutics/targeted therapeutics, radiotherapy, photodynamic therapy and certain oncolytic viruses can induce a form of cancer cell death called ICD which is accompanied by spatiotemporally defined emission of danger signals. These danger signals along with other factors help cancer cells undergoing ICD to activate host innate immune cells, which in turn activate T cell-based immunity that helps eradicate live (or residual) surviving cancer cells. The emergence of ICD has been marred by some controversy. ICD has been criticized to be either experimental model or setting-specific or mostly a concept based on rodent studies that may have very limited implications for clinical application. However, in recent times it has emerged (through mainly retrospective or prognostic studies) that ICD can work in various human clinical settings hinting towards clinical applicability of ICD. However a widespread consensus on this issue is still transitional. In the current Research Topic we aimed to organize and intensify a discussion that strives to bring together the academic and clinical research community in order to provide a background to the current state-of-the-art in ICD associated bench-side research and to initiate fruitful discussions on present and future prospects of ICD translating towards the clinical, bedside reality.

Abstract

This book includes updated information about microRNA regulation, for example, in the fields of circular RNAs, multiomics analysis, biomarkers and oncogenes. The variety of topics included in this book reaffirms the extent to which microRNA regulation affects biological processes. Although microRNAs are not translated to proteins, their importance for biological processes is not less than proteins. An understanding of their roles in various biological processes is critical to understanding gene function in these biological processes. Although non-coding RNAs other than microRNAs have recently come under investigation, microRNA still remains the front runner as the subject of genetic and biological studies. In reading the collection of papers, readers can grasp the most updated information regarding microRNA regulation, which will continue to be an important topic in genetics and biology.

Abstract

As the Century of Biology begins to bear fruit, through the translation of predictive biological understanding into applications that enhance the human condition and maintain biodiversity, the almost infinite potential of marine biological resources will be unlocked. Although Marine Biotechnology already has delivered products for medicine, food, bioenergy, nanomaterials, and bioremediation, less than 5% of our vast oceanic environment has been explored. Marine Biotechnology is a scientifically and economically expanding enterprise that is poised to harness the enormous but uncharted functional diversity of marine life, with its novel and rich array of biodesigns and biosynthetic capabilities. From this pursuit comes new genes, chemicals, materials, and inspirations for the benefit of industry, nutrition, and medicine, and which enable the sustainable use and management of the world’s oceans.
