Abstract

Anopheles mosquitoes are highly important insects due to their involvement in the transmission of human malaria and its devastating consequences in endemic countries worldwide. In 2010 alone, malaria was responsible for an estimated 660,000 deaths. As the study of Anopheles species and populations is a key element for reaching the goal of malaria elimination, an enormous amount of information has accumulated over the past century, and together in recent decades with the advent of novel technologies the acquisition of new knowledge has accelerated even further. The originality of this book is to offer the latest compilation on various research, new concepts, paradigms and innovative approaches for the control of anophelines using state-of-the-art methodologies and analysis. Abstract

Foodborne disease like salmonellosis and toxoplasmosis are amongst the most significant causes of hospitalization in the U.S. and globally. Gastrointestinal infections alter gut microbiomes and increase permeability to toxins. Various invasions by microbial, fungal, viral and parasitic agents stimulate inflammation, a defensive mechanism of the body’s immune system. Other stimuli include environmental stimuli, oxidative stress, aging and the physiological process. A long-lasting, persistent and excessive inflammatory response is a significant risk factor for developing various chronic inflammatory and infectious diseases.Different nutritional and dietary life styles, whether poor or lacking essential nutritional elements, as well as excess intake, can result in inflammatory complications and loss of function. Nutritional deficiency is linked with several infectious and inflammatory diseases as a cause or consequence. For instance, protein deficiency was reported in orphanages to provoke microbial and fungal complications including Pneumocystis pneumonia. Similarly, protein deficiency is a tell-tale sign of several parasitic diseases. Studies indicate that nutrients, such as amino acids, oligosaccharides, and short-chain fatty acids exert inhibitory and anti-inflammatory functions. These investigations help to understand nutritional contributions to the prevention, treatment and taming of certain inflammatory and infectious diseases. Infectious and inflammatory complications go hand-in-hand with malnutrition.The purpose of this Special Issue is to publish related new, basic and translational findings and clinical trials in this area. Other investigations or review articles were sought to link infectious and inflammatory diseases with nutrients. In addition, novel diagnostic, preventive and therapeutic modalities were invited to aid the development of nutritional strategies for the treatment and/or prevention of inflammation and infection. Original reviews were of particular interest to advance our understanding of signaling pathways, and the molecular and biochemical mechanisms behind the effects of nutrients on inflammatory and infectious diseases.

Abstract

In this chapter, we review knowledge gained so far on mosquitoes from An. moucheti and closely related species, as well as the An. nili complex. We highlight specific bionomical, ecological and genetic attributes that distinguish these species from the most well-known major African malaria vectors, providing opportunities for further research on neglected aspects of vector biology and control.

Abstract

Zoonoses are currently considered as one of the most important threats for public health worldwide. Zoonoses can be defined as any disease or infection that is naturally transmissible from vertebrate or invertebrate animals to humans and vice-versa. Approximately 75% of recently emerging infectious diseases affecting humans are diseases of animal origin; approximately 60% of all human pathogens are zoonotic. All types of potential pathogenic agents, including viruses, parasites, bacteria and fungi, can cause these zoonotic infections. From the wide range of potential vectors of zoonoses, insects are probably those of major significance due to their abundance, high plasticity and adaptability to different kinds of pathogens, high degrees of synanthropism in several groups and difficulties to apply effective programs of population control. Although ticks, flies, cockroaches, bugs and fleas are excellent insects capable to transmit viruses, parasites and bacteria, undoubtedly mosquitoes are the most important disease vectors. Mosquito borne diseases like malaria, dengue, equine encephalitis, West Nile, Mayaro or Chikungunya are zoonoses with increasing incidence in last years in tropical and temperate countries. Vertebrates can also transmit serious zoonoses, highlighting the role of some carnivorous animals in rabies dissemination or the spread of rodent borne diseases in several rural and urban areas. Moreover, the significance of other food borne zoonoses such as taeniasis, trichinellosis or toxoplasmosis may not been underestimated. According to WHO, FAO and OIE guidelines an emerging zoonotic disease can be defined as a zoonosis that is newly recognized or newly evolved, or that has occurred previously but shows an increase of incidence or expansion in geographical, host or vector range. There are many factors that can provoke or accelerate the emergence of zoonoses, such as environmental changes, habitat modifications, variations of human and animal demography, pathogens and vectors anomalous mobilization related with human practices and globalization, deterioration of the strategies of vector control or changes in pathogen genetics. To reduce public health risks from zoonoses is absolutely necessary to acquire an integrative perspective that includes the study of the complexity of interactions among humans, animals and environment in order to be able to fight against these issues of primary interest for human health. In any case, although zoonoses represent significant public health threats, many of them still remain as neglected diseases and consequently are not prioritized by some health international organisms.

Abstract

Genome-wide association studies (GWAS) for complex disorders with large case-control populations have been performed on hundreds of traits in more than 1200 published studies (http://www.genome.gov/gwastudies/) but the variants detected by GWAS account for little of the heritability of these traits, leading to an increasing interest in using family based designs. While GWAS studies are designed to find common variants with low to moderate attributable risks, family based studies are expected to find rare variants with high attributable risk. Because family-based designs can better control both genetic and environmental background, this study design is robust to heterogeneity and population stratification. Moreover, in family-based analysis, the background genetic variation can be modeled to control the residual variance which could increase the power to identify disease associated rare variants. Analysis of families can also help us gain knowledge about disease transmission and inheritance patterns. Although a family-based design has the advantage of being robust to false positives, novel and powerful methods to analyze families in genetic epidemiology continue to be needed, especially for the interaction between genetic and environmental factors associated with disease. Moreover, with the rapid development of sequencing technology, advances in approaches to the design and analysis of sequencing data in families are also greatly needed. The 11 articles in this book all introduce new methodology and, using family data, substantial new findings are presented in the areas of infectious diseases, diabetes, eye traits, autism spectrum disorder and prostate cancer.

Abstract

According to the World Health Organization (WHO), in 2012 infectious diseases and related conditions account for more than 70% of premature deaths across 22 African countries and estimated 450 000 people worldwide developed multi-drug resistant tuberculosis. This alarming situation, of great public health concern, calls for the urgent development of novel and efficient responding strategies. The employment of important research platforms, such as genomics and proteomics, has contributed significant insight into the mechanisms underlying microbial infection and microbe-host interaction. In this Frontiers Research Topic, we aim to produce a timely and pertinent discussion regarding the current status of “Proteomics of microbial Human pathogens” and the role of proteomics in combating the challenges posed by microbial infection and indeed acquired anti-microbial resistance. As the field of proteomics progressed from 2-DE gel based approaches to modern LC-MS/MS based workflows, remarkable advances have been reported in terms of data quantity and quality. Given the immediate and enormous advantages that high resolution and accurate mass spectrometers have brought to the field, proteomics has now evolved into a robust platform capable of generating large amounts of comprehensive data comparable to that reported previously in genomics studies. For example, detection of the complete yeast proteome has been reported and other small proteomes, such as those of bacteria, are within reach. Mass spectrometry-based proteomics has become an essential tool for biologists and biochemists, and is now considered by many as an essential component of modern structural biology. Additionally, the introduction of high-resolution mass spectrometers has driven the development of various different strategies aimed at accurate quantification of absolute and relative amount of protein(s) of interest. Emerging targeted mass spectrometry methodologies such as; Selected Reaction Monitoring (SRM), Parallel Reaction Monitoring (PRM) and SWATH, are perhaps the latest breakthrough within the proteomics community. Indeed, through a label free approach, targeted mass spectrometry offers an unequalled capability to characterize and quantify a specific set of proteins reproducibility, in any biological sample. Usefully, Aebersold and colleagues have recently generated and validated a number of assays to quantify 97% of the 4,012 annotated Mycobacterium tuberculosis (Mtb) proteins by SRM. As such, the Mtb Proteome library represents a valuable experimental resource that will undoubtedly bring new insight to the complex life cycle of Mtb. Finally, as reviewed recently in Frontiers Research Topic, mass spectrometry-based proteomics has had a tremendous impact on our current understanding of post translational modification (PTM) in bacteria including the key role of PTMs during interaction of pathogenic bacteria and host interactions. We believe that our understanding of microbial Human pathogens has benefited enormously from both 2-DE gel and modern LC-MS/MS based proteomics. It is our wish to produce an integrated discussion surrounding this topic to highlight the existing synergy between these research fields. We envisage this Research Topic as a window to expert opinions and perspectives on the realistic practicalities of proteomics as an important tool to address healthcare problems caused by microbial pathogens.
