Abstract

Metabolic syndrome (MetS) can be considered as a clustering of several risk factors such as obesity, hypertension, insulin resistance and dyslipidemia, which could lead to the development of diabetes and cardiovascular diseases (CVD). There are several underlying causes for MetS including overweight, physical inactivity and genetic factors. However, the underlying mechanisms that leads to MetS are still poorly understood. Therefore, the aim of this E-book is to provide a space where researchers holding different backgrounds could shed some light onto the pathophysiology of different risk factors involved in MetS, mostly from translational research worldwide.

Abstract

Obesity and its co-morbidities, including atherosclerosis, insulin resistance and diabetes, are a world-wide epidemic. Inflammatory immune responses in metabolic tissues have emerged as a universal feature of these metabolic disorders. While initial work highlighted the contribution of macrophages to tissue inflammation and insulin resistance, recent studies demonstrate that cells of the adaptive immune compartment, including T and B lymphocytes and dendritic cells also participate in obesity-induced pathogenesis of these conditions. However, the molecular and cellular pathways by which the innate and adaptive branches of immunity control tissue and systemic metabolism remain poorly understood. To engage in growth and activation, cells need to increase their biomass and replicate their genome. This process presents a substantial bioenergetic challenge: growing and activated cells must increase ATP production and acquire or synthesize raw materials, including lipids, proteins and nucleic acids. To do so, they actively reprogram their intracellular metabolism from catabolic mitochondrial oxidative phosphorylation to glycolysis and other anabolic pathways. This metabolic reprogramming is under the control of specific signal transduction pathways whose underlying molecular mechanisms and relevance to physiology and disease are subject of considerable current interest and under intense study. Recent reports have elucidated the physiological role of metabolic reprogramming in macrophage and T cell activation and differentiation, B- and dendritic cell biology, as well as in the crosstalk of immune cells with endothelial and stem cells. It is also becoming increasingly evident that alterations of metabolic pathways play a major role in the pathogenesis of chronic inflammatory disorders. Due to the scientific distance between immunologists and experts in metabolism (e.g., clinicians and biochemists), however, there has been limited cross-talk between these communities. This collection of articles aims at promoting such cross-talk and accelerating discoveries in the emerging field of immunometabolism.

Abstract

This new book entitled "Non-Alcoholic Fatty Liver Disease Research 2016" covers a selection of recent research topics and current review articles in the field of nonalcoholic fatty liver disease (NAFLD) that have been recently published in a monographic Special Issue of the International Journal of Molecular Sciences (IJMS) journal. NAFLD is an “umbrella” definition that encompasses a spectrum of histopathological liver changes ranging from simple steatosis to nonalcoholic steatohepatitis (NASH) with/without fibrosis, “cryptogenic” cirrhosis and hepatocellular carcinoma. NAFLD has become one of the most common forms of chronic liver disease worldwide and its prevalence is expected to continue rising. It is now becoming increasingly clear that the clinical and economic burden of NAFLD does not only affect the liver, but also affects the risk of developing cardiovascular disease, type 2 diabetes mellitus, colo-rectal neoplasms, chronic kidney disease and other extra-hepatic diseases that have a considerable impact on health-care expenditures. Against this background, further research is needed to better understand the natural history, and the molecular pathogenesis of NAFLD, as well as to elucidate the underlying mechanisms by which NAFLD contributes to the increased cardiometabolic risk, and to disclose novel and effective treatment strategies for this increasingly prevalent and burdensome disease.Some of the leading international researchers in this area expressed a willingness to contribute to this book providing an updated, state-of-the-art view on the aforementioned topics, and also suggesting novel research avenues for NAFLD.

Abstract

During the last decade, a high volume of work has been published on the health-promoting effects of berries (e.g., blueberries, cranberries, blackberries, etc.) that are rich in antioxidant phytochemicals, polyphenols. Consuming a diet rich in polyphenols has been documented to attenuate the risk of chronic diseases, such as cardiovascular disease, certain cancers, diabetes mellitus, and neurodegenerative disorders. Recent evidence also reveals that the biological effects of polyphenols extend beyond their traditional antioxidant role.This Special Issue includes 10 peer-reviewed papers, including original research papers and reviews. They present the most recent advances in the role of berry antioxidants, not only in maintaining health but also in preventing and/or reversing disease both in cell culture, animal models and in humans. Additionally, the molecular mechanisms and signaling pathways modulated by berry antioxidants are presented. Chapters include the role of berry antioxidants in whole fruit and leaves on the metabolic syndrome, obesity, diabetes and glucose intolerance, cancer, inflammation, oxidative stress and neuroprotection as well as cardiovascular disease. Abstract

Cadmium (Cd) is an environmental toxicant of continuing public health concern worldwide, because total diet studies have shown that Cd is present in virtually all foodstuffs. Consequently, foods that are frequently consumed in large quantities, such as rice, potatoes, wheat, leafy salad vegetables, and other cereal crops, are the most significant dietary Cd sources. Moreover, Cd has chemical propensities that confer the potential to interfere with the physiological functions of calcium and zinc. Evidence of a wide range of diverse, toxic effects of Cd is increasingly apparent. In this collection, environmental Cd exposure is linked to an increased risk of chronic kidney disease that is known to be a cause of morbidity and mortality worldwide. Cd is also implicated in an early onset of menarche and deaths from cancer, especially in the uterus, kidney, and urinary tract. Moreover, Cd-induced kidney injury is replicated in Sprague Dawley rats, as is Cd-induced periodontal disease. Experimental studies suggest that the development of kidneys in fetuses and the function of insulin-producing cells may be adversely affected by Cd and that metformin, an anti-diabetic drug, is ineffective in Cd-intoxicated Wistar rats.

Abstract

In recent years, great attention has been paid to polyphenols due to their positive effects on health. One of the most widely-studied phenolic compounds is resveratrol. This molecule, which is naturally present in some foods, shows beneficial effects on various physiological and biochemical processes, thus representing a potential tool for the prevention or the treatment of diseases highly prevalent in our society. Several of these beneficial effects have been observed in human beings, but others only in pre-clinical studies so far, and therefore, it is mandatory to continue with the scientific research in this field. Indeed, new knowledge concerning these issues could enable the development of novel functional foods or nutraceuticals, incorporating resveratrol, suitable for preventing or treating diseases such as cancer, cardiovascular diseases, obesity, dislipemia, insulin resistance and diabetes, liver diseases, etc.

Abstract

Food insecurity is a complex ‘wicked’ problem that results from a range of unstable and uncertain physical, social, cultural and economic factors that limits access to nutritious food. Globally, 800 million people are under-nourished, and around 2 billion are overweight/obese or have micronutrient deficiency. These populations are largely positioned in developing countries where disease burden is high and impacts health budgets and productivity. Similarly developed countries, cities and neighbourhoods are experiencing a greater emergence of vulnerable populations. This is in part explained by the change in the food production and manufacturing, the retraction in economic climates, the increase in food price, and in some regions reduced food availability and access.Vulnerable groups include but are not limited to migrant populations, Indigenous people, elderly, pregnant women, those with disability, homeless, young children and youth. Poor nutrition at significant periods of growth and development and during life impact long term health outcomes increasing non-communicable disease prevalence, health cost and reducing economic productivity.
