Abstract

Human tissue and biobank research is of increasing importance for understanding the causes of widespread diseases and developing effective therapies. However, while the success of biobank research depends on the availability of a large number of samples and the consolidation of collections across country borders is very desirable from the perspective of researchers, the legal and ethical requirements for the procurement, storage and use of human tissue samples are rather heterogeneous across different countries. Moreover, the lack of comprehensive supranational regulation on human tissue and biobanking can be seen as posing a serious threat to transnational biomedical research. Against this background, it was one of the aims of the EU-funded Tiss.EU project (“Evaluation of Legislation and Related Guidelines on the Procurement, Storage and Transfer of Human Tissues and Cells in the European Union – an Evidence-Based Impact Analysis”) to analyse the ethical and legal regulation of human tissue and biobank research across the 27 European Member States plus Switzerland. The results of nine international workshops and three conferences are gathered in this volume. While the country reports evaluate the implementation of ethical and legal guidelines at a national level, point out their strengths and deficits, and, where required, create an evidence base for the revision of said legislation, the conference reports address more general ethical and legal issues in this field. The volume is completed by a final presentation of project’s results.

Abstract

Tissue engineering, the creation of human tissue in the laboratory by the use and propagation of human cells to develop commercial products, faces many problems concerning its legal regulation. There have been numerous alterations to the laws governing biotechnology –derived medicinal products. However, difficulties remain regarding their legal classification. This work shall give a legal survey of the whole procedure of tissue engineering concerning German and European law.

Abstract

Tissue engineering emerged back in the 1990s as a new concept to overcome the problem of tissue and organ failure. Over recent decades, there has been incredible progress towards the regeneration of tissues such as bone, heart valves, cartilage, cornea, and retina. In terms of ocular tissue engineering, despite the scientific and strategic incentive for reconstructing ocular tissues, there is also a tremendous need for novel therapeutic options in treating numerous eye diseases related to tissue failure. The aim of this Special Issue is to discuss tissue engineering applications of ocular tissues including but not limited to cornea, retina, and lenses.

Abstract

The concept of regenerating tissues, with properties and functions that mimic natural tissues, has attracted significant attention in recent years. It provides potential solutions for treating many diseases and other healthcare issues. To fully realize the potential of the approach, it is crucial to have a rational biomaterial design to create novel scaffolds, and other materials systems suitable for tissue engineering, repair and regeneration. Research advances on the topic include the design of new biomaterials and their composites, the scaffold fabrication via subtractive and additive manufacturing approaches, the development of implantable scaffolds for disease monitoring, diagnostics, and treatment, as well as the understanding of cells–biomaterial scaffolds interaction. This Special Issue, “Novel Biomaterials for Tissue Engineering”, covers a selection of timely research activities in the field of biomaterials for tissue engineering and regeneration purposes. Promising findings on different approaches to design and develop new biomaterials, biomaterial systems and methods for tissue engineering, are presented and discussed. Recent advances in biofabrication techniques for tissue engineering are additionally demonstrated. The issue comprises a series of state-of-the-art experimental works, up-to-date review articles and commentaries.

Abstract

Soft tissue and bone sarcomas disproportionately affect children, adolescents, and young adults. Although considered a rare disease, sarcomas continue to have a devastating effect on these patients and their loved ones, and their impact on our society far exceed their relatively low prevalence. Because of its rarity and heterogeneity, clinical decision making on management of sarcomas are often individualized and without a consensus treatment strategy. Furthermore, research to improve clinical outcomes are particularly challenging as sizeable studies are hard to produce. Abstract

The Tohoku University Graduate School of Dentistry first introduced the concept of “Interface Oral Health Science”, designed to establish and maintain healthy oral cavities, which are home to a number of mixed systems. Included in those systems are: (1) host tissues such as teeth, mucosa, muscle and bone, (2) parasites and microorganisms cohabiting the surfaces of the oral cavity and (3) biomaterials that are used for the rehabilitation of oral functions. In addition, (4) these systems are subject to severe and complex mechanical forces. Therefore, it is critical to promote dental studies that integrate a wide range of interdisciplinary research as medicine, agriculture, material science, engineering, and pharmacology. With this incentive, international symposiums for interface oral health science have been held several times in the past. The concept has since refined and expanded, the result being the “Biosis-Abiosis Intelligent Interface,” and projects aiming at the creation of highly functional and autonomic intelligent interfaces are ongoing. This book brings together a number of studies on incentives and projects by leading authors. Topics include biosis-abiosis interface of dental implants, biomaterials in interface science, biomedical engineering interface and cell manipulation and tissue regeneration. Readers not only from the field of dentistry but also many related areas will find this book a valuable resource.

Abstract

The development and function of the immune system is dependent on interactions between haematopoietic cells and non-hematopoietic stromal cells. The non-hematopoietic stromal cells create the microenvironment in which the immune system operates, providing an architectural landscape for hematopoietic cell-cell interactions and molecular cues governing haematopoietic cell positioning, growth and survival. Not surprisingly, therefore, aberrant stromal cell function has recently been shown to play a key role in the development of disease pathologies associated with immune dysfunction. For example, remodelling of lymphoid tissue stroma and the development of ectopic tertiary lymphoid tissues are characteristic of many infectious and inflammatory diseases and stromal cells have a recognised role in lymphoma and tumour development and resistance to therapy. An increased understanding of the molecular basis of stromal cell differentiation and function in these varied contexts will provide new tools to promote research on stromal cell biology and immune dysfunction, and potential new targets for therapeutic intervention in diseases with a major impact on public health. The importance of stromal cells and the molecular mechanisms of stromal cell function in the regulation of immune responses have only recently been appreciated and thus represent an exciting new area in immunology.

Abstract

The biological outcome of Hyaluronan (also hyaluronic acid, abbreviated HA) interaction with its CD44 or RHAMM receptors recently attracted much attention within the scientific community owing to a Nature article by Tian X et al. (Nature 2013; 499:346-9). The article described a life span exceeding 30 years in naked mole rats, whereas the maximal lifespan of mice, to which the naked mole rat is related, is only 4 years. This observation is accompanied by the finding that the naked mole rat, in contrast to the mouse, does not develop spontaneous tumors during this exceptional longevity. The article provides evidence that interaction of long tissue HA (6000-12,000 kDa) of the naked mole rat with cell surface CD44, in contrast to the interaction of short tissue HA (less than 3000 kDa) with the mouse CD44, makes the difference. More specifically, this communication shows that the interaction of short HA with fibroblasts’ CD44 imposes on them susceptibility for malignant transformation, whereas the corresponding interaction with long HA imposes on the fibroblasts a resistance to malignant transformation. The article does not explain the mechanism that underlines these findings. However, the articles, that will be published in the proposed Research Topic in the Inflammation section of Frontiers in Immunology, can bridge not only this gap, but also may explain why interaction between short HA and cell surface CD44 (or RHAMM, an additional HA receptor) enhances the development of inflammatory and malignant diseases. Furthermore, the articles included in the proposed Frontiers Research Topic will show that cancer cells and inflammatory cells share several properties related to the interaction between short HA and cell surface CD44 and/or RHAMM. These shared properties include: 1. Support of cell migration, which allows tumor metastasis and accumulation of inflammatory cells at the inflammation site; 2. Delivery of intracellular signaling, which leads to cell survival of either cancer cells or inflammatory cells; 3. Delivery of intracellular signaling, which activates cell replication and population expansion of either cancer cells or inflammatory cells; and 4. Binding of growth factors to cell surface CD44 of cancer cells or inflammatory cells (i.e., the growth factors) and their presentation to cells with cognate receptors (endothelial cells, fibroblasts), leading to pro-malignant or pro-inflammatory activities. Going back to the naked mole rat story, we may conclude from the proposed articles of this Frontiers Research Topic that the long HA, which displays anti-malignant effect, interferes with the above described pro-malignant potential of the short HA (perhaps by competing on the same CD44 receptor). Extrapolating this concept to Inflammation, the same mechanism (competition?) may be valid for inflammatory (and autoimmune) activities. If this is the case, long HA may be used for therapy of both malignant and inflammatory diseases. Moreover, targeting the interaction between short HA and CD44 (e.g. by anti-CD44 blocking antibodies) may display also a therapeutic effect on both malignant and inflammatory diseases, an issue that encourages not only fruitful exchange of views, but also practical experimental collaboration.

Abstract

Abstract

Catechols are widely found in nature taking part in a variety of biological functions, ranging from the aqueous adhesion of marine organisms to the storage of transition metal ions. Catechol-based natural and biomimetic molecules have thus been the focus of intense research aimed at mimicking and translating this nature-inspired chemistry into new functional materials and systems with tailored properties. The aim of this book is to present a concise survey of the state-of-the-art of basic and applied research on bioinspired catechol systems.
